Dermatophyte Test Medium

Dermatophyte Test Medium (DTM) Sterile.
This media  is specifically for Dermatophyte fungi.

The composition of the DTM medium (g / liter):

- Peptone = 10 g
- Glucose = 10 g
- Cycloheximide = 0,5 g
- Gentamicin sulphate = 0,1 g
- Chlortetracycline = 0,1 g
- Phenol red = 0,2 g
- Agar-agar = 17 g

Note :

DTM medium containing:

1. Cycloheximide functions as an inhibitor for fungal contaminants.
2. Gentamicin and Chlortetracycline function as an anti bacterial.
3. There is a pH indicator phenol red which at acidic pH, the phenol red is yellow. This leads to a yellow medium (A). While at alkaline pH, phenol red is red. This leads to a red medium (B).

B. DTM Positive

  • Dermatophyte fungi begin to grow so then the DTM color change from yellow to red.
  • If the fungus is a Dermatophyte, and the metabolites of the fungus are alkaline substances, so the medium becomes red.
  • With the color change from yellow to red we can surmise that the fungus is a Dermatophyte.
  • Dermatophyte fungi are usually identified by the typical formation of macroconidia and microconidia, after 2 weeks culture.
